What it is
SLU-PP-332 is a small molecule, not a peptide. It is a synthetic pan-agonist of the estrogen-related receptors ERR alpha, beta, and gamma, with the highest potency at ERR alpha. It was developed in academic medicinal chemistry laboratories and appears in preclinical exercise-mimetic and metabolic research.
How it works
The ERRs are nuclear receptors that govern mitochondrial biogenesis and oxidative metabolism gene programmes in muscle. SLU-PP-332 is proposed to switch on an acute aerobic exercise gene programme through ERR alpha. In the published mouse work, ERR alpha activity was required for the endurance effect.
Regulatory status
- Cosmetic ingredient
- Research compound
- In clinical trials
- Approved medicine
Not approved as a medicine by the FDA or the EMA. Some are registered in individual countries. Supplied for laboratory research only.
What published research is studying
- Running endurance and oxidative muscle fibre type in mice
- Energy expenditure and fatty acid oxidation in mouse models of obesity
- Fat mass accumulation and insulin sensitivity in metabolic syndrome models
- ERR pharmacology, chemical optimisation, and detection of related analogues
Key findings
In mice given SLU-PP-332 twice daily for 7 days before testing, running time was about 70% longer and distance about 45% further than in vehicle controls.
Billon C et al., ACS Chem Biol 2023The same study reported an increase in type IIa oxidative skeletal muscle fibres, and showed the endurance effect depended on ERR alpha activation.
Billon C et al., ACS Chem Biol 2023In diet-induced obese mice dosed for 28 days, fat mass gain was nearly abolished versus vehicle, with about 12% body weight loss and higher resting energy expenditure.

What the studies measured
Study durations
- Mouse endurance study: 7 days of dosing before exercise testing, and a separate 2-week arm for grip strength (Billon et al. 2023).
- Diet-induced obesity study in mice: 28 days of dosing; the ob/ob arm ran 12 days (Billon et al. 2024).
Endpoints
- Treadmill running time and distance in mice
- Skeletal muscle fibre type and oxidative gene expression
- Fat mass and body weight by NMR body composition
- Resting energy expenditure and respiratory exchange ratio
- Fasting glucose, insulin, and glucose tolerance
How strong is the evidence?
There are no published human trials of SLU-PP-332. Every reported effect comes from mouse and cell studies, and its pharmacokinetics in humans are unknown.
