What it is
CJC-1295 with DAC is a long-acting analog of growth hormone releasing hormone. It is a tetrasubstituted form of human GRF(1-29) carrying a maleimidopropionamide group, the Drug Affinity Complex, which bonds covalently to cysteine 34 of circulating albumin. It appears in endocrinology research on sustained stimulation of the growth hormone and IGF-I axis.
How it works
It binds the GHRH receptor on pituitary somatotroph cells, prompting growth hormone synthesis and release. Albumin conjugation is thought to shield the peptide from renal clearance and from dipeptidyl peptidase-4, which extends its measured plasma half-life to several days.
Regulatory status
- Cosmetic ingredient
- Research compound
- In clinical trials
- Approved medicine
Not approved as a medicine by the FDA or the EMA. Some are registered in individual countries. Supplied for laboratory research only.
What published research is studying
- Pharmacokinetics and duration of growth hormone and IGF-I elevation in healthy adults
- Whether pulsatile growth hormone secretion is preserved during continuous GHRH receptor stimulation
- Growth restoration in GHRH-deficient animal models
- Serum protein markers of growth hormone and IGF-I action
Key findings
In two randomized, double-blind ascending-dose trials in healthy adults, a single subcutaneous dose raised mean plasma growth hormone 2- to 10-fold for 6 days or more. Mean IGF-I rose 1.5- to 3-fold for 9 to 11 days.
Teichman SL et al., J Clin Endocrinol Metab 2006In the same trials the estimated half-life was 5.8 to 8.1 days. After repeat dosing, mean IGF-I stayed above baseline for up to 28 days, and no serious adverse reactions were reported.
Teichman SL et al., J Clin Endocrinol Metab 2006In healthy men aged 20 to 40 years, trough growth hormone was 7.5-fold higher one week after a single injection. Mean growth hormone rose 46% and IGF-I rose 45%, with pulse frequency and amplitude unchanged.
Ionescu M, Frohman LA, J Clin Endocrinol Metab 2006In GHRH knockout mice treated for 5 weeks from 1 week of age, once-daily CJC-1295 produced normal body weight and length. Longer intervals of 48 and 72 hours did not fully normalize growth.

What the studies measured
Study durations
- Ascending-dose trials in healthy adults: 28 days and 49 days (Teichman 2006).
- Growth hormone pulsatility study: 12-hour overnight sampling before and one week after a single injection (Ionescu 2006).
- GHRH knockout mouse growth study: 5 weeks of treatment (Alba 2006).
Endpoints
- Peak growth hormone concentration and growth hormone area under the curve
- Serum IGF-I over time
- Plasma half-life and standard pharmacokinetic parameters
- Growth hormone pulse frequency, amplitude and trough level
- Body weight, body length and femur or tibia length in mice
How strong is the evidence?
Human evidence is limited to small early-phase pharmacokinetic and pharmacodynamic studies in healthy volunteers, with no published phase 3 outcome trial.
